Mucin disc preparation for analysis of muco adhesive strength??

Dear all, I need to prepare mucin discs following reported procedure (mucin=250 mg, compression force = 10 tonnes for 30 seconds), but I am not getting disc rather getting aggregates.

can I add any moistening/binding agent??? If yes than how much???
