I want to know "how can I filter / remove the blood and mucin from the media containing bacteria, mucin, and blood".
The purpose of my research is to analyze the microbial profile in the three kinds of media.
Media containing bacteria only
Media containing bacteria and mucin
Media containing bacteria and blood
I cultured the media for about 5 days, and then I extracted DNA from this media and performed 16S PCR & gel electrophoresis.
While there was a definite band of the sample that derived from media containing bacteria only, no band appeared from the sample that derived from media containing bacteria and mucin.
After I realized that the mucin can inhibit PCR, I want to know how to filter / remove the mucin from the media containing bacteria and mucin.
Also, there was a weak band of the sample that derived from media containing bacteria and blood.
So, I want to filter / remove the blood from the media containing bacteria and blood.
I tried to filter the blood from the media. When I used 5um filter, no filtration effect was observed (RBCs penetrated through the 5um filter).
When I used 1.2um filter, nothing penetrated through the 1.2um filter.
So, I want to know how to remove/filter the blood and mucin from the media.
