I am doing some cell based assay. I am culturing cells in 25 cm2 cell culture flask. now for my assay I need to seed 1X10^5 cells/well in 24 well cell culture plate. Now, how can I calculate the volume of medium (containing cells) need to be taken to seed 1X10^5 cells per well? And for all wells what will be the total volume of medium required to seed the said amount of cell?
Is there any formula?

Resuspend the cells in 3-4mL of medium and do the total cell count per mL (say X). Divide the total cell count (X) with ur required count (say Y, for u it is1X10^5) and u'll get the total culture volume (say Z). Make up the total culture volume upto Z and u'll get ur desired count.
e.g. Cells suspended in 4 mL of culture media.
Total cell count (X) = 1X10^6 cells/mL
Required cell count (Y) = 1X10^5 cells/mL
Total culture volume (Z) = 1X10^6 / 1X10^5 = 10
Add 6 mL to initial cell suspension and Make up the culture volume to 10mL. Each mL will give u 1X10^5 cells/mL. Seed the wells of 24-well plate with 1mL.
