Thanks a lot everybody. Today I made a test, I transfected myoblast with GFP in following conditions: 4X10^4 or 6x10^4 cell/well (plate 24 well), with Lipofectamine 2000, L3000, Fugene and PEI . I intend to see the transfection efficiency of these transfections reagents after 24, 48 and 72h, because our idea is save money and get could results, because the price of PEI is really good than the anothers.
Today I fixed the group of cells of 48h of transfection and tomorrow will be the last one group. I have some experiments running together, but I intend to have a look on microscopy this week. As soon as possible I intend share my experience with this test.

PS: This PEI is commercially (polysciences ), I got an alicot from neighbour lab for a test, if I have sucess, I intend to buy it.
Another option is Promofectin, this is what I use in the lab. However best transfection efficiency up to 80% if you want to transfect high numbers of cells is the Amaxa Nucleofector Kit by Lonza. I recently tested this for the first time and its really good.
I have infected them with Adenovirus and the efficiency is very high..but now I want to try overexpressing a gene using Lipofectamine 2000...if anybody has tried can you give me the protocol and tell how the efficiency is?
I am also interested in comparison of lipofectamin and PEI effectivity in C2C12, but I cannot find your results. would you be so nice and share it again?
