Does anyone know if 25 or 50 uM of chloroquine treatment could be toxic for the cells (C2C12 cells or any other cell line) in a time-course treatment like 1, 3, 24, and 48 h?
This is coming rather late. I hope some others can benefit. As far as I know,
25 or 50 uM of chloroquine for short time course is fine but its use for long time especially 48hrs could be toxic to cells. However, its toxicity is also cell dependent as certain cells (liver cells) are able to metabolise the drug while others can not. Chloroquine itself has been shown to have a relatively long half life so all these can be put into consideration when planning your experiment.
