The tissue was embbeded in Spurr Resin. The infiltration process was 1:3, 1:2, 1:1, 2:1, Pure (Resin x Acetone). When I went to make the cut, I coudn't cut nothing. Is there any protocol to follow to re-embbeding a plant tissue that not infiltrated? How can I remove the previous resin?
Just prepare new samples from the first step and increase infiltration time. If the samples are extremely important and you don't have more tissue, it is possible to cut them of from the blocks with a sharp blade under a preparative microscope. But definitely there will be serious artifacts and this way doesn't look so good.
