I have not tried doing ELISPOT without ethanol pre-treatment. I am afraid of not getting high-quality spots. The ELISPOT plate with PVDF membrane is recommended for good quality results. PVDF membranes are naturally hydrophobic. Pre-treatment with ethanol will turn it into hydrophilic. Only a hydrophilic membrane can combine enough capture antibodies effectively, and enough quantity of capture antibodies will give good quality spots.
So, pre-treating the ELISPOT plate with 35% ethanol for 30 secs is an important step.
Prewetting of PVDF is essential. Throw a slice into water and see what does (not) happen. You also may use other short chain alcohols like Methanol. Propanols should work, too.
