I did an MTT assay with 2 drugs (Piroxicam and Sulfameter) when I got my results for the 12,24,48,72 hours of incubation with my drug at the following concentrations [ug/ml] (128,64,32,18,8,4,2,1,0.5) all the results were 3.5 when looking at absorbance. I followed my protocol to the last letter, but all my results remin 3.5, I am not sure what to do.
It may be due to contamination. Or, the drug may have been insoluble in the solvent and formed a precipitate, and after treatment, it didn't show its effect.
It appears that Piroxicam does not exhibit significant cytotoxicity in certain cell lines.
- 10.2131/jts.44.701, there is no cytotoxic effect up to 1500 µg/ml in liver cells.
- 10.1016/j.colsurfb.2011.11.050, Piroxicam in microspheres shows no cytotoxic effect up to 300 µg/ml in pancreatic cells.
I expect that Sulfameter behaves similarly.
Could it be that you tested concentrations that were too low?
Did you have a negative control with no drug? Also one with just the diluent? Just looking with your eyes, did all wells look the same color? If the answer to all is yes, I would try to increase the dose.
