Is it right to leave tissue biopsy in the freezer 24 hrs with or with out media then estimate the cancer stem cells after 24 hrs.?

we receive tissue at 5 pm for cancer stem cell estimation 2nd day 10 am , is it better to leave the tissue at 4 C or -20C ?

Hello Ibrahim.

If you determine CSCs by FACS, store tissues in Hank's solution (cold at 4C) with minimum of added NaN3 to Hank's solution. Wash tissues in cold Hank's solution+NaN3 before storing at 4C.

we have no sorter so we digest tissues then add specific CDs. then incubate then run on flowcytometry. We depend upon the specificity of different CDs to bind with the receptors of required CSCs.
