Is it right to leave tissue biopsy in the freezer 24 hrs with or with out media then estimate the cancer stem cells after 24 hrs.?

we receive tissue at 5 pm for cancer stem cell estimation 2nd day 10 am , is it better to leave the tissue at 4 C or -20C ?

Thank you very much for your last answer,but is there any cross reactivity for specific CDs between CSCs and any other cells ?

we have no sorter so we digest tissues then add specific CDs. then incubate then run on flowcytometry. We depend upon the specificity of different CDs to bind with the receptors of required CSCs.
