When i do RNA isolation many would suggest that RNA isolation have to be done using refrigerated centrifuge.
Is it really necessary? What if i use a regular centrifuge and works at room temp. without air conditioning? Will those conditions affect my RNA quality? Will the condition differs between Trizol protocol and spin column protocol?
Temperature is a form of energy. As it reduces rate of chemical reactions also reduce. Hence freezing and thawing / frequent temperature changes would affect the quality of any biomolecules. Now you may guess the answers.
To check the RNA degradation it is best to use refrigeratored centrifuge.

But, in case you lack refrigerated centrifuge, you can proceed ahead with regular centrifuge. This will reduce your quantity but still you will get RNA that can be used for further processing. If you have lab centrifuge, you can try placing it in refrigerator during RNA isolation.

How about RNA isolation with column? Is it behave similarly even when the RNA is binded to column?
Yes Dylan Lawrie . Still my answer is suitable here as well (Temperature is a .....).
But using column will give better results.
