My team working with microspore culture, we are usually using NLN medium in liquid form. Since we need to add active charcoal, we try solidify NLN medium but it cant be solid. May you have good reason for this? Thank you before
did you check the pH of the medium? its quite likely that it is a bit too acidic for the medium to solidify using agar at generally recommended (0.8%) concentraion.
Base on the data of the base medium from HiMedia (see link) it is quite acidic pH~4.1, so you need to adjust it.
I have made culture media for fungi with pH 4.0 with 2% agar with no problem but with a lower concentration you'll probably have a noticeable hydrolisis.
If you use gellan gum (aka Gelrite or Phytagel) you may need to increase the amount of divalent ions, e.g. Ca2+, as it gelification is ion-dependent.
