We are culturing MIN6 cells in our lab. And we use complete medium with %10 FBS, which is heat inactivated according to protocols. But I would like to try to use non-heat inactivated FBS. Is it necessary for these cell lines? Cause every protocol that I see refers to using heat-inactivated FBS.
We have used Hyclone Characterized FBS which I believe IS heat inactivated, but if I were you, I'd split off a flask and just try non HI serum in one flask and compare the the results you get with your flask that is cultured with heat inactivated serum. That is the best way to ensure that your results are not compromised by the switch. It could be that some experiments are not affected but others are. Bets is always to see what happens in your hands.

I have tried that. And there are no visible differences between DMEM included heat inactivated FBS or active FBS.
