I isolate exosomes from condition medium according to ultracentrifugation protocols.
after 3 centrifugation (10 min x300g) then (15min x2000g) then (30 min x10,000g) , medium collected and filter (0.2um) before 120min x100,000g centrifugation.
if I forget to filter before last centrifugation?

You need not worry. The use of 0.2um filter before ultracentrifugation is just to clarify the conditioned medium. It won't affect your end result.

You might have other entities in your pellet besides exosomes. We have identified an extremely small group of cells with a size range of 0.1 to 2.0 microns with the same differentiation potential that can co-localize with exosomes. We have termed them adult-derived totipotent stem cells for their ability to form ALL somatic cells of the body, gametes, and the nucleus pulposus of the intervertebral disc (the only adult derivative of the notochord. There are actually two subsets. The first is sized at 0.1 to 1 micron and grows in suspension. The second is sized from 1 to 2 microns and grows on a type-I collagen substratum (it does not grow in suspension). Please see attached papers.
