Anybody could explain to me which method is better for measurement the level of toll like receptor protein in tumor tissue , Immunohistochemistry vs western blot?
IHC will tell you where it is, or which cells are expressing it, but is not great at telling you how much is there. Western blotting is at least semi-quantitative so you can figure out how much TLR4 there is compared to control.
I have not specifically worked with TLR4 before. If you're still not sure, you can always try both methods and see how they compare. Your success will depend heavily on how good your antibody is.
I agree with Joshua said. i have small experience in TLR4. to measure more details, WB is better way. But western blot can not identified where is the TLR4 located. Immunohistochemistry is the best way to know the TLR4 expression site in the tissue. even if its not as precision as WB, we can quantified the expression level by image analysis. you can use imageJ, Fiji or adobe to measure the percentage of TLR expression and compare with the normal tissue
I agree with the above advice. However, you should also consider the accessibility of positive and negative control for your experiment. I suggest you to select a method which you have verified control samples for it.
