Infos I have on the sheet is 23.3 nmoles, OD 5.20. The question is, is it already 100mM or do I need to dilute? No idea! Please let me know if you have experience with this!
Not sure where did you buy your primers from, usually I purchase them from Invitrogen. They always give you the option of buying your primers dry or liquid. The last means the primers are already dissolved in ultra pure water. The standard concentration is 100 micromolar. But they should have give you a datasheet specifying all the primer details. if not i would suggest give them a call and they will give you the details. Do you have one vial for forward sequence and one vial for reverse sequence?
Regarding the shipment, I think it is quite common to ship without cooling. At least, my primers always come in an envelope and without refrigeration. However place them in -20 as soon as possible. Also, I always prepare alliquots of my forward and reverse primers in water, so I do not freeze-thaw the vials constantly.
Don`t worry about your primers!! it would be working , although it depends on your primer length and preparation methodology. For diluting your primers, it is preferred to adjust the concentration to 100 picomoles per microliter (equals to 100 micromolar). You have to add TE or ultrapure distilled water to you primer solution to a final volume of 233 microliters. you will then have a 100 micromolar solution of your primer. Good luck
Thank you all! it was so helpful to discuss with you really. I realize it was 100 micromolar liquid and it came without dry ice only within envelope! so I freezed directly .
