I am working with HPV pseudovirions and after some silver staining and ELISA experiments I realized I only have few micrograms of capsid protein (L1)/ml in my preps.
Pseudovirions preps contain 33-39% iodixanol. I need to diafiltrate/concentrate these preps and I was wondering how.
My colleague works with HPV VLPs (containing cesium chloride) and usually makes a fast dialysis + amicon tubes diafiltration, so I was thinking to try the same. However, I am not sure about the compatibility of iodixanol (Optiprep) and dialysis membranes or amicon filters. I will try her strategy on a VLP sample containing 39% iodixanol and check by SDS-PAGE coomassie staining if I lose some protein during the steps.
...But maybe someone has experience with concentration/diafiltration of HPV pseudovirions and can tell me their strategy?
