Hi everybody! I forgot to add FBS 10% to EMEM medium and the cells grew well, died very less. After freezing them and put them to grow again, I still didn't add the FBS and they stopped to grow: very small, circular even if the % of dead cells was low.
If I now make new medium EMEM + 10% FBS or 15% FBS can they survive and grow? Have you ever experienced that? Many thanks!!

It is always recommended to use 10 % FBS for HepG2 cells. Please stick to that. Regarding your second question (Growth issue after freezing and simulteneous sub culturing again): Probably there is a mycoplasma contamination due to which there is a sluggish or no growth!!..Go for Mycoplasma contamination test!!

The lack of FBS might also be the reason for growth issues. Serum is a source of amino acids, carbohydrates and vitamins - as well as growth factors. Especially FBS is known for its growth-promoting properties.
