Does anybody know of, or have a method suitable for HPLC analysis that allows to separate violaxanthin from neoxanthin? I want to quantify the xanthophyll cycle (violaxanthin, antheraxanthin and zeaxanthin) and I used a standard for these three compounds but not neoxanthin. Any experience / advice / recommendations for dealing with such HPLC technique would be very welcome.

Accurate, precise, selective and, importantly, selective as well as absolute quantification, currently, can be carried out only by means of mass spectrometry; however, within the framework of our (mine and my co-author's ones as has been indicated in the authorship of the works) innovative stochastic dynamic concept and model formulas applicable to quantify exactly the mass spectrometric variable 'intensity'.
Our method is tested so far at mixtures of structurally very similar analytes at pg.(mL)-1 and ng.(L)-1 concentration levels, showing coefficients of linear correlations between our formulas and the experiment within /r/ = 0.99997 - 1.
Please, consider in this context, the content of works [1-3]. The chemometric analysis of six replicates of carbohydrate mixtures in work [3] shows /r/ = 1.

The error contribution to the mean area value, due to deconvolution processing of a pattern can be evaluated looking at reference [1*], herein. The fact that the latter reference deals with IR-patterns does not deceive the evaluation, because of it is a mathematical data-processing with corresponding chemometric treatment of the outcomes.
The corresponding error contribution to chromatographic data can be viewed looking at references [1,2] in my previous posting, as well.
By means of chromatography there is unable to achieve superior method performances, due to these reasons, even examining single chromatographic peak.
