We have always used Probond (Invitrogen) but Promega resin is cheaper. I was wondering how the Promega resin performs, to see if it is worth to change brands. Thank you!!
Have you tried WorkBeads 40 Ni-NTA? It has a capacity 50-70 mg/ml resin (of course protein-to-protein dependent) and has good price. Also available precharged with other metal ions (Co, Zn and Cu) if you want to check different selectivities. Using cobolt ions instead of Ni may give you a more pure His-tagged protein, but you can lose some yield. If you are working with eucaryotic expression systems which usually include reducing and/or chelating reagents in the cell culture media you should use WorkBeads NiMAC instead which is tolerant towards these additives and the nickel ions will not be stripped off from the resin during loading.
