I have limited access to the FACS and was hoping there is a way to do both tests in as little time as possible to avoid loosing viability. I will eventually move on to more sensitive cells so time will be even more important... I can provide details if necessary, but didn't want to make the question too specific since the main problem is the possible tests! Thanks!
Not knowing al the details, one possible quick way is to use a microscope and imaging. Use the fluorescent filter to measure your transfection efficiency and then you can also do a trypan blue exclusion (or other similar ideas, eg. dapi/7aad etc) using normal brightfield (or other fluorescent filters) and superimpose the images (and you can add pseudo-coloring as well). Of course, the numbers will be limited by field size but it's a start.
