I read a paper using lipo2000 to co-transfect cas9 mRNA and sgRNA, but I failed when using lipo3000. I followed Thermo fisher's protocol (without using P3000), the cell line was HepG2.2.15. Do I need to try again with P3000, or is there anything I didn't notice? I just want to use lipo3000 as a positive control to prove my sgRNA is intact, because the new deliver tech sometimes can be unstable, I want to be sure at least sgRNA is ok.
sgRNA is modified with CY5, so I can clearly read the transfect efficacy.
