I tried the CTAB protocol and a salting out protocol for DNA extraction from mature leaves of a tree. The supernatant is in both cases dark green and I suppose that DNA solution still contains also other organic compounds. Does anyone know how to overcome this problem in order to get a clearer supernatant and improve my extraction? I have to work with big amounts (grams of leaves) so I am not using kits. Thanks.
