Hi all, I have SEM images of electrospun nanofibers and I'd like to measure its porosity/diameter. However I'm not sure how to do it with ImageJ or any other software. Can anybody help me please? Thanks in advance
Use the line ROI tool to draw a line across your scale bar which is saved on your SEM image.
Go to 'analyze --> Set Scale'
Put in the distance of your scale bar. (say, 10 microns)
Use the measurement tools and 'Analyze --> Measure' to measure the distance.
It is also possible (if you have good contrasts, etc), to get ImageJ to measure many pores automatically and give a histogram.
This depends on 3 things 1) Resolution of your microscope 2) Magnitude of your pores 3) Clear images where you can see all the porous structures one by one
Or
Method 2. you can install the "DiameterJ" plugin. It calculates pores, fiber diameter and orientation.
If you spin PLA in CHCl3 you might have fibers big enough for nanoCT scanners. Then you have the real scaffold pore sizes rather then 2D ones.
Or
Method 3. For porosity measurement you can go for liquid displacement method or BET rather than image analysis.
Hello dear, you can measure porosity by uploading image in Autocad program and then specifying the distance between the boundaries of each porous by using roller tool then the number will be shown. Finally to get exact unit numbers you must convert image scale to the same scale in SEM image, then compare exactly with shown number of porosity diameter.
