Hi guys,
Currently, we are culturing primary cancer cells. They look good at P1 to P3, but senescent quickly later passages. So, I am wondering how those commercial cancer cell lines were established, such as Hep G2, U251, MCF7. Any protein factors were transfected in?

There are some ways to immortalize the cells (like introduction SV40 virus or mutations to primary cells or activating telomerase activity), but in general believe, cancer cells grow continuously. Therefore you do not need that kind of immortalization. You have to optimize your culture conditions and medium supplements (like extra growth factors etc.). Because your culture is heterogeneous.

These commercials cell lines even many others , are derived from patient tissues. Those patients were carrying cetain specific mutations which these cells naturally presents. Moreover, they were transfected with with several other insertion, deletion, overexpression, knockout like events used to be done for getting permanent characteristics and makes it establised, immortal cell lines. The perfect cells were selected, clonal selection followed with mass culture or explant culture and then goes for commercial production.
