I am curious to know much plasma i need for exosome isolation by ultracentrifugation method for miRNA and LC-MS/MS work,
is it 2 ml plasma is okay for this work ?

The answer to this question very much depends on the specific isolation protocol used in the experiments.
I can't comment on the LC-MS/MS part, but for miRNA characterization (small RNA-Seq), we successfully isolated EVs from as little as 1 ml serum by UC.
1ml if you are doing NGS for validation
300-500 ul if you are doing rt-qPCR .
