Hi, I'm starting to do bisulfite sequencing, and I'm the first person to do this experiment in my lab so I need some advices.

From numerous protocols and papers, when doing cloning-based bisulfite sequencing, it says selecting minimum 10 colonies for sequencing is needed to get statistically meaningful result.

I have a question in this step, what if there's so many positive colonies?

I mean, if I get over 100 positive colonies, is selecting 10 colonies sufficient to get meaningful result?

Or do I need to select more colonies? If not, is there any method I could get meaningful result?

I've searched for many protocols, but nobody explains about this.
