We use hippocampal organotypic slices (prepared P5-6) for electrophysiology and other applications. In our lab it has been customary to prepare slice culture media, aliquot and store at 4C for up to two weeks. Lately we are using fewer slices each week and I have a significant amount of leftover media after two weeks. However, I feel that if I reduced the total volume of media prepared I wouldn't be able to accurately measure some of the components of the media. Does the media last longer than two weeks? Are there any labs using organotypic slices for electrophysiology that treat their slice culture media differently?
To our experience, medium for hippocampal slices does not last more than two weeks, it's critical for electrophysological recordings. I recommend to make less amount and use it before two weeks.
HI, if the media is sterilized by filtration it could be stored for weeks at 4°C. But this is not a case if you use some unstable component like pyruvate or ascorbic acid. Ususaly unstable components are added prior the experiment
We have used media that are over a month old, stored at 4 degrees C. We have not noticed any difference. We use a different medium for dissection/slice preparation and for slice maintenance. We often keep the former frozen at -20 indefinitely, and the latter at 4 degrees. We have not explored the outer limits of how long these can be stored, but we generally use them until they are used up, which sometimes is over a month.
