We use hippocampal organotypic slices (prepared P5-6) for electrophysiology and other applications. In our lab it has been customary to prepare slice culture media, aliquot and store at 4C for up to two weeks. Lately we are using fewer slices each week and I have a significant amount of leftover media after two weeks. However, I feel that if I reduced the total volume of media prepared I wouldn't be able to accurately measure some of the components of the media. Does the media last longer than two weeks? Are there any labs using organotypic slices for electrophysiology that treat their slice culture media differently?
