Hello everyone, I have a question about my research topic. I used a tyrosine kinase inhibitor (Dasatinib) to improve detection of CD8+ cells, according to a few publications (Wooldridge et al. 2009, Dolton et al. 2018). Now I'm trying to understand how it works because it is claimed that Dasatinib inhibits downregulation of T cell receptor and CD8 molecules. Does anyone know how it works on a molecular level? Thanks!

TCR/CD8 expression levels are determined by the rate constants for synthesis, endocytosis, recycling, and degradation.
In resting T cells, the TCR/CD8 slowly and constitutively cycles between the plasma membrane and the intracellular compartment; TCR triggering/signalling induces an enhancement in the endocytic rate constant leading to TCR/CD8 down-regulation. Hence, tyrosine kinase inhibitor (Dasatinib) stablize and enhance TCR/CD8 amount on cell surface.
