Hi Sarath! I always use a surfactant like polysorbate 20 to 80 (Tween) to break the surface tension caused by the hydrophobins. I get much more conidial units per ml and, in the case of tween I didn't ever see any difference in the viability of the conidia. Just a droplet in one liter will be enough. You can also pipette 100 microliters, but it is hard, due to the viscosity. The best thing: you can autoclave it once it is in water solution. Good luck!

I don't fully understand. It seems like you have petri dishes (agarized culture, but then you say you have 10 ml. is it liquid culture or conidial suspension. In any case this should help:
