I am working in field of artificial insemination which depends mainly on the presence of semen with high sperm motility. However, estimation of the sperm motility is subjective and differs from one to another. So, how can sperm motility be estimated accurately by implementing a new technique of computer-based-system.

you can use software like Quali sperm, CASA, Auto sperm , MMC Sperm: Computer Aided Sperm Analyzer - MMCSoft and SCA® CASA System for automated semen analysis ...etc.

CASA (Computer Aided Sperm Analysis) is becoming more reliable. It consists of a microscope mounted camera attached to a computer with dedicated software. The software allows for setting specific limits w.r.t. size etc in order to accurately identify the spermatozoa and track them. These tracks are then analysed to provide accurate information regarding various kinematic parameters (e.g. motility, progressive motility, and velocity such as VSL, VCL, LIN, ALH ....and hyperactivation). Depending on the software that you buuy, you can also perform morphology, viability and DNA fragmentation analysis.
I think the options have been well covered by others. You say you are working in AI but I am unsure whether you mean in humans or animal species. Different machines and software work more or less effectively with different species so you need to think about how that affects your options. Secondly many systems measure a whole host of sperm motility parameters many of which are of marginal relevance if you main interest if % progressively motile is your main interest. Of course cost of machines arises with their sophistication so make sure you are not purchasing a Ferrari when all you want is a Ford!

Only to give you some additional information. In my opinion, when you use CASA systems you have to take into account that if you want to get "accurate" measurements, you should invert additional time to manually reevaluate samples (video captures) in order to avoid sub- or overstimated results (that could be time consuming), i.e. either debris selected as immobile spermatozoa, or segmented pathways that are selected as various spermatozoa. You also have to take into account the sample support method. I recommend you a Makler chamber.

I carry out studies based on motility determination. I can understood you because it is difficult to determine subjective motility in similar samples. However, computer assisted systems is expensive and it is not warranty to determine accurate motility. I think if you have enough experience on subjective motility you can determine accurate motility as well as CASA. Nevertheless, CASA has some advantages compared with conventional analyses. You can determine velocity and other kinematic properties of spermatozoa with CASA. However, you can determine hyperactivation in spermatozoa without CASA.
CASA systems simply has two fundamental units. One is hardware containing Phase-Contrast Microscope, C-mount, camera and computer. Another one is software containing program that records images from microscope and analyses sperm motility and kinematic motions. CASA can determine curvilinear, straightline and average path velocity of spermatozoon (VCL, VSL, VAP micrometer per second) and also head beat per second. In this way, you can reach more accurate results.
CASA needs chamber systems instead of slip and coverslipe. And it is disposable and you have to buy for each analyse.
We started a new project in 2013 and we have developed a novel computer assisted sperm analyses system. It is cheaper and it does not need chamber system. However it is prototype at the moment but I hope in a few year we will have developed commercial version.
