Are the techniques named as MFM (Magnetic force Microscopy), CD (Circular Dichroism), PPMS (Physical Property measurement system) sufficient to find out this particular information or are there any kinetic studies also involved in this particular area?
You can deduce a Langmuir saturation adsorption capacity curve and this will give you the protein adsorbed value you are looking for.

You can quantify the protein in your starting solution and the supernatant recovered after coating. By mass balance you can actually determine the amount coated to your particles.

but i think that will give me an ambiguous interpretation as i am not sure whether it is free protein or unbound protein after the reaction has taken place.

The number of amino groups on your particles can be determined by a spectrophotometric assay ( for exemple based on a fluorigenic reaction of phthaldialdehyde (OPA) with primary amines.
Another method than fluorescence is to determine the surface area of particles by a low
temperature nitrogen adsorption method ( this method is called BET ) You fill your magnetic particles (3 mg) into a capillary tube, and you look for nitrogen adsorption and desorption were carried out at 77 K. You can measure then the free surface area by the BET method ( but you need like mg of NPs)
You have also the SEM microscopy ( more precise than AFM on spherical NP)

that was a wonderful suggestion....however i will be going with fluorescence spectroscopy, CD and TGA. But the concern is the physical state of the characterized sample is liquid. then ill it be possible to go ahead ?
About fluorescence microscopy - you can trace/visualize many proteins by using it but quantitative studies will be difficult
I would recommend ATR-FTIR measurements. if you can produce a thin film of you nanoparticles, you will get information about the amount of proteins adsorbed and their conformation in situ and in real time.
