The Ganoderma boninense in the lab was contaminated with other fungi (which is white in color as well).
I tried to subculture into different media but the contamination is still there.
I would like to know what type of methods can be use to clean up the fungus contamination of the Ganoderma boninense. Thanks a lot.
by that you mean getting rid of other contaminating organisms, e.g. Tricodermas? or do you mean a specific product?
Try transferring hyphal tips. You might be able to get rid of it.
Hyphal tipping and water agar (or both) are great suggestions. If that doesn't work you could try a selective medium: Worrall JJ. 1991. Media for selective isolation of hymenomycetes. Mycologia 83(3):296-302.
Hi Dr. Hale, I don't know contaminated with what kind of fungus yet but under the microscope it looks like aspergillus.

Do the hyphal tipping under a stereo dissecting scope, around 100+ magnification. It is easiest to see the hyphae if you use transmitted light. The scopes with a swivel mirror below the stage work best. Shine the light on the mirror and angle it so that the hyphae are highlighted on a relatively dark field.
Of course you want a very fine tool. One option is a thin, double-sided razor blade. With pliers you can snap a small piece of the edge of at an angle so there is a point. Then you need a handle to put it into, like a scalpel handle.
Water agar is good for this because the hyphae are less dense, and you can work on a single one. Only problem is you may be hyphal tipping the contaminant. That's where selective media are handy.
It just occurred to me that if you have a good quality scope and can zoom in, you may be able to see the clamp connections on the Ganoderma hyphae. That way you can distinguish them from the hyphae of the contaminant.
