Hi, I have done the extraction from the culture filtrate of fungi using hexane. So currently I'm having the hexane extracts and I developed it using the preparative thin layer chromatography (PLC). I am planning to extract the fractions from the PLC. But my concern is that, what kind of solvent should I use to extract the compounds from the silica of PLC? Can I use just hexane? Or ethyl acetate? Or both to completely extract the compounds from each fractions? Thanks.
this is depend on solubility of your sampling extracted . so you can use ethyl acetate, ether or other available solvent then you can evaporate the solvent under vacuum to dry the sample.
To dissolve the compounds and extract it from silica, it is better to use the same solvent in which you extracted the fractions..In this case it should be hexane rather than ethylacetate.
If you followed hot extraction you should use both solvents in equal proportion but if it was cold extraction, I think hexane is enough.
Thank you very much for the advice. But for the extraction, if I extract with hexane, followed by ethyl acetate should be fine? I actually dissolved my hexane extract in ethyl acetate. I don't know whether I do it this way is it correct or not.
scrape the band, dissolve in th same solvent used in the PTLC, filter off the silica gel, and evaporate using water bath or rotor vaporator. There you have your sample
