We are culturing HepG2 cells in low attachment wells to make them grow in shpheroid forms in RPMI 10% SVF. Then T cells are added to the wells and the culture medium becomes 50% RPMI 10% SVF and 50% Gibco Optimizer (specially designed for T cell culture but with unknown composition).
Desired results: HepG2 spheroids grow at the same rate with or without T cells.
Actual result. HepG2 shperoids basically stop growing and go towards slow death, but grow very well when T cells are present.
It is like HepG2 cells use T cells as feeder cells. Mind that T cells are fine too, it is not like HepG2 are killing them to use as food. Is this something that is possible for HepG2 cells? Or is it something in the optimizer that bothers them (the experiment to compare HepG2 spheroid growth in different culture mediums has not yet been done in my lab, but will be shortly).
