I am thinking in using this expression system to express dihydropteridine reductase. I want to know if anyone has used this system before and what advantages and disadvantages they have over other expression systems.
I have used this vector before. It works very well. It really depends on what sort of protein you want to express. Its recommended to express the protein at 15C but I have seen it working well at 18C as well. Really depends on the protein.
Hi, I used pCold-TF. This vector is really good. The TF stands for Trigger-Factor and it is used as a solubility tag for your target protein. Obviously, you can cleave it.
The advantage of using these pCold vectors is that the promoter is fully active at low temperature. The effect is then double : first the low temperature prevents the aggregation and misfolding of your protein and second: despite the low temperature the rate of expression is really high thanks to the tuned promoter.
