%100 confluent MEF feeder cells in 6-well culture dish got shrinked and showed a phenotype looks like under a serious stress condition after we changed the medium from MEF medium (including FBS) to IPSC medium (including KSOR).I wonder if anyone has seen such a toxic effect of knockout serum replacement on MEF feeder cells.
Never. Are you sure that is KSR? On which passage you have MEF? In my culture MEF looks like...... usually after 5th passage, so maybe this is the reason.
Have you try on other cells? If your KSR is toxic, it will kill other fibroblast too....

I have not noticed any toxic effect of KOSR medium on MEFS.
However, the morphology of the MEFs do change from relatively flat cells (in FDMEM) to elongated cells, when changed to KOSR medium.

MEF cells, as you'd know, require FBS/FCS for their proliferation. When we introduce them to stem cell environment and their media, including KOSR, the MEFs change morphology and might not look very happy. That's why, ideally, we need to split the stem cells every 3-4 days to transfer them to fresher MEFs. It is normal if you have had MEFs in culture with stem cells and specific media, that MEFs would start looking different and unhealthy.
I wouldn't call it a toxic effect of KOSR, but just the absence of serum in the media, which they require to stay healthy. If you could share some pics, it might be more helpful..
