According to the protocol, I don't see many problems, if we look at the standard sample, the bands are satisfactorily separated. But usually when we have a high sample concentration, the band becomes more "full", making it difficult to determine its size. If you decrease the concentration of your sample it is very likely that the band will become more evident. And a better resolution photo would also help to evaluate better.
