I would like to ask whether someone has experienced that bone marrow derived MDCSs would stimulate splenocyte proliferation instead pf suppressing it? I performed a co-culture of BM-MDSCs and CFSE labelled mouse splenocytes in 1:4, 1:10, 1:20, 1:40 ratio. Splenocyte were activated by ConA (1-2.5 ug/ml). After 3 days of co-culturing I see the opposite of expected result. I get more proliferating T cells at 1:4 ratio than at 1:40, in other words, if I have more MDSCs in the culture they stimulate the splenocyte proliferation instead of suppression. I have tried to different concentration of ConA 0.5, 1, 2.5 ug/ml but I see similar results.
I would appreciate if anyone could comment on this!
