I am using the kit Genomic DNA from soil from Macherey-Nagel but I do not have DNA. I follow the manual instructions. The soil is a mixture od organic soil:sand:zeolite (1:1:1). Anyone has suggestions and/or possible explanations for what is happen? Anyone use this kit?
Haven't used the kit. Though my suggestion would be to try different amounts of sample to optimize. You could be adding too much sample or too little sample. Also try vortex for longer times to shear those particles/clumps and release cells.

The MN kit you use for the DNA is good enough for the purpose, but the problem might be your sample. As those samples has more amount of inert material, it's often difficult to get the DNA out. I would avoid increasing the time of bead beating or vortexing as it will surely degrade and shear the DNA. You can treat your samples (quantity according to manual) with proteinase K to lyse the cells and follow the MN protocol. Hence you could get good quality of DNA in sufficient amount to work with.
