I want to perform MTT assay to prove application of a certain drug for biomedical application in general (not oral dose), can I use any mammalian cell lines?
Basically, yes you can in order to determine any acute toxicity. However, it is much more useful to choose relevant cell lines depending on your route of administration e.g. if you plan to deliver the particles via the direct inhalation then an airway cell line would provide relevant data. In general it is best to choose more than one cell line if possible as different cell types will react differently to particles. A good starting point would be lung, liver (if I had to pick one) and kidney cell lines since these organs are where the majority of your particles will end up.
