Has anyone experienced an increase in the absorbance reading with increasing concentration of a substance with expected antioxidant effect using the DPPH assay?
First, before use whatever technique you must understand the chemistry/physics behind the technique. The product of antioxidant oxidation might have a strong light absorbance in the same wavelength as DPPH.
In normally DPPH assay when we increase the concentration of the compound or crude extract, increase the absorption due to increase the formation of Diphenylpicrylhydrazine (nonradical). If your sample has a colour you have to reduce the absorbance in your sample (every concentration).
