I am trying to do an imaging experiment where I need to observe cellular organelles. I do not have poly L lysine in my lab. My question is whether this line grow on coverslip with out coating?
I think the SH-SY5Y cell lines have mixed culture properties (adherent and suspension). It is looking difficult that they grow on cover slip without coating. You can try in 4 or 8 wells chamber slide, but in case of suspension cells, you have to fix them on the slide using some suitable method.
Hello Dipanwita, The SH-SY5Y cell lines have the property to adhere the non tissue culture treated surfaces. You should get proper adherent cell on a coverslip.
If not, try the following-
irradiate your coverslip with UV for 10-15 minutes in laminar hood before seeding.
You can increase the serum concentration also if they are still not attaced.
For my experiments I used Fibronectin (Sigma-Aldrich) to treat surfaces, then whashed twice with dpbs before to add the cells; it helped a lot to improve cell adhesion.
