MTT method
We have used 540nm for MTT reading and I think it doesn't meter which type of cells are you using.
I also used 540 nm for MTT assay and this assay is not cell line specific.
540 or 560 nM is the most efficient,
i always substract a 720nM readout as a background control.
We work with HeLa cells and read absorbance at 570 nm for MTT. It is not cell line specific as they have said.
Its the wavelength where the colour is giving the best od values.
I use LPS at 5ug/mL as a positive control in the Griess method, but nitrite production is very low.
