I would like to prepare N2B27 medium for ESC culture, and according to the protocol I am going to obtain 1L of medium. Does anyone know how long can I keep it in the fridge or if it is possible to aliquot the medium and freeze it at -20ºC? Any advice about conservation of the medium will be kindly apreciated.

Generally the recombinant proteins (FGFs, TGFs etc) and growth supplements (eg N2, B27 etc) are kept as small aliquots and added to the basal media on the day of use and also in the volume required. Differentiation media (complete media) can be stored a week at 4deg Celsius, but I would advice you to prepare your complete medium as an when required Preparing 1L of complete media is not advisable, as you cannot change any concentration if required, aliquoting the supplements would help. Hi Marta, I usually aliqoute B27, FGF, EGF and store them in -20. They are only good for 2 weeks. You can calculate how much media you would use in a 2-week period and just make up what you needed. Do not aliquote and freeze your complete medium. Good luck!

N2B27 is good for 1 week in the fridge and protected from light! Once made up. You can make aliquots and store at -20. Once thawed, again, it is good for 1 week in the fridge in the dark. Hope that helps.
