Dear all.
I am about to start B cell immortalization experiments with EBV, but the vast majority of my blood samples come from frozen whole blood in EDTA tubes and without any cryopreservation product (like DMSO 10%). I know that during the thaw, cells are likely to die and it might be nearly impossible to have a good efficiency when culturing PBMCs from these samples. Thus, I would like to know if someone has tried to culture PBMCs from frozen blood or if someone has the same problem as me and they could give me some advice to face this hitch.

It's quite impossible to get even few live cells from frozen (uncryopreservation) blood. I tried few times from frozen PBMCs but it didn't work.
If somehow you get to know any working protocol for this, do let me know. It will be very helpful as we have precious patient samples.
