I want to add glucosylceramide to cells. I redissolved the lipid obtained from Avanti Polar Lipids in absolute ethanol to obtain a 100 mM solution. From this I took 4 ul in PBS and sonicated 30 min a sonicator water bath. The solution stayed cloudy also after heating to 37 degrees and an additional sonication of 30 min.
I have no experience, does the lipid/PBS mixture have to become clear after sonication or is it normal that it stays cloudy? How can I be sure that the micell formation during the sonication was successful?
