We determine monoglycerides by normal phase HPLC using a ELS detector obtaining reliable results. Now we have a new equipment (SHIMADZU UFLC) with the same type of detector. However, when implementing the same technique for monoglycerides we are just obtain a flat response. The solvent Used is a mixture of hexane and 2-propanol. Everything in the equipment seems to be working fine, since we can obtain nice chromatographs for other compounds (sterols). We already clean de nebulaizer And everything inside the oven.

The answer is no. But as I recall monoglycerides and monosaccharides have to be derivatized for GC (TMS as I recall) because they are polar or water loving. Since this is normal phase (no water in the mobile phase) you will probably have to perform the same.
Thanks Bruce, but for MG analisis by HPLC they do not need to be derivatized. We are using a silica based column, thus during the initial ellution (with hexane and 2% 2-propanol) the monoglycerides must get attached to the functional groups of the silica. Then after changin the solvent polarity by increasing the proportion of propanol, the MG’s must get released and, therefore, must run along with the solvent.
