Hello,

I am doing a drug release analysis, placing sample in a dialysis membrane, placing membrane in a flask with water and removing water samples for analysis via hplc.

The method that I have found in jounral articles is via method 1 below.

Method 1. Sink maintained: e.g. remove 1mL sample for analysis, replace with 1mL

Method 2. Complete buffer replacement: e.g. remove 1mL sample for analysis, discard buffer and replace with fresh buffer completely.

I am using Paclitaxel which is hydrophobic and has very low solubility in water, so it makes sense (to me) to do the second way so the buffer doesnt get overconcentrated...

I am wondering if there is any reason why I shouldnt do method 2?
