How do I get rid of the background signal using the Bio-Rad Quantity One?

I've coated plastic water bottles (PET) with Nile Red to detect the plastic degradation by Ideonella sakaiensis. When using the settings for Sypro Ruby, the coated and uncoated plastic looks the same. I would like my uncoated plastic not to light up. How do I eliminate the background signal using the Bio-Rad Quantity One?
